Anyone have tips on removing a Virus Manually From Your Computer?

I know there's a lot of great software, but what if that doesn't work? Is there a way to manually remove a virus?

Personally, I know there is, but I also know it's a PITA and not for those who don't know what they're doing.

Anyone out there have any experience with removing a virus manually from their PC?
